Macromolecular crowding develops heterogeneous environments of gene expression in picoliter droplets

چکیده
Drawing of the PDMS-based microfluidic chip. Two inlets including one for the oil and one for the in vitro transcription and translation mix (IVTT). Droplets are generated using PDMS based microfluidic chips, the IVTT mixture is pinched off at regular intervals by oil generating monodisperse water in oil emulsions. The droplets are stored in the storage chamber and can be monitored over time with fluorescence microscopy. Plasmid map of pRSET-CFP/ YFP (see Methods section for more details).

Enhanced transcription rates in membrane-free protocells formed by coacervation of cell lysate.
Liquid-liquid phase transitions in complex mixtures of proteins and other molecules produce crowded compartments supporting in vitro transcription and translation. We developed a method based on picoliter water-in-oil droplets to induce coacervation in Escherichia coli cell lysate and follow gene expression under crowded and noncrowded conditions. Coacervation creates an artificial cell-like en...
متن کاملCell-Free Protein Expression under Macromolecular Crowding Conditions
BACKGROUND Cell-free protein expression (CFPE) comprised of in vitro transcription and translation is currently manipulated in relatively dilute solutions, in which the macromolecular crowding effects present in living cells are largely ignored. This may not only affect the efficiency of protein synthesis in vitro, but also limit our understanding of the functions and interactions of biomolecul...
متن کاملMacromolecular Crowding Regulates the Gene Expression Profile by Limiting Diffusion
We seek to elucidate the role of macromolecular crowding in transcription and translation. It is well known that stochasticity in gene expression can lead to differential gene expression and heterogeneity in a cell population. Recent experimental observations by Tan et al. have improved our understanding of the functional role of macromolecular crowding. It can be inferred from their observatio...
متن کاملExcluded-Volume Effects in Living Cells**
Biomolecules evolve and function in densely crowded and highly heterogeneous cellular environments. Such conditions are often mimicked in the test tube by the addition of artificial macromolecular crowding agents. Still, it is unclear if such cosolutes indeed reflect the physicochemical properties of the cellular environment as the in-cell crowding effect has not yet been quantified. We have de...
متن کاملThe Role of Crowded Physiological Environments in Prion and Prion-like Protein Aggregation
Prion diseases and prion-like protein misfolding diseases are related to the accumulation of abnormal aggregates of the normal host proteins including prion proteins and Tau protein. These proteins possess self-templating and transmissible characteristics. The crowded physiological environments where the aggregation of these amyloidogenic proteins takes place can be imitated in vitro by the add...
